In a template you only define the general layout of the item, not the list of items. You can create an event and use that to copy the items from or create a Content document in the Sourcing Library. Support can assist if you have any issues.
Its not a best practice to have too many variables in your event content so to reduce some you may choose to create weighted average of the quantity slabs based on the historical data or approximate projection say for the next quarter/year.
If you do that you should be able to come up with a cost factor that you can build into your event at the line level. You may then choose to go ahead and create the event with 200 line items that are all combinations of 20 source to 10 destinations. The Quantity slabs can then go into the line item as additional cost terms with default price for a single trip. The weighted average should be incorporated with the use of the common cost factor mentioned earlier.
Needless to say this has to be done at the Project/Event level and not at the template level unless u need like 100 such projects to be executed every year.

There are couple of options:
A) Matrix of say 20 Origin (From Locations) to say 10 Destination (To Locations) - create it as a line item
B) Quantity Slabs say < 100 kg / 101-500 kg / 501 - 1000 kg / 1001 kg - 5000 kgs - create it as Matrix
or you may look into alternative bidding template.
